The probabilities of problems and their causes may be estimated or may be calculated from historical data accumulated by use of the flow chart, or some combination of the two. These probabilities are incorporated into the answer cells of the dependency matrix. The resulting flowchart is tested and evaluated, and the results of testing and evaluation are used to make modifications in the matrix of questions and answers. The flowchart is then made available for execution by a problem determination engine, usable by staff at a help center or directly as self-help to a customer over a network. Experience from use of the problem determination engine is accumulated for use in further refinements of the dependency matrix and its probabilities. | 09-18-2008 |

20130315879 | METHOD FOR EXPANSION OF STEM CELLS AND THE USE OF SUCH CELLS - The present invention demonstrates that SALL4A and SALL4B are strong positive regulators of hematopoetic stem cell expansion. HSCs receiving expression of SALL4A or SALL4B are able to achieve a high-level of expansion. Cultures of SALL4-transduced cells results in extensive HSC expansion with over 1000-fold higher levels than controls within 2 to 3 weeks and expanded HSCs show no or very little maturation. Moreover, the expansion occurs quite rapidly with significant HSC growth in just a few days. In addition, SALL4-induced HSC expansion exhibits no impairment of hematopoietic cell differentiation. SALL4 appears to function in the maintenance of an undifferentiated proliferation state and block cell differentiation for HSCs. | 11-28-2013 |
20150342999 | METHODS AND COMPOSITIONS FOR TREATING DIABETES WITH IPS DERIVED PANCREATIC BETA-LIKE CELLS - Diabetes mellitus is characterized by either the inability to produce insulin (Type 1 diabetes) and or as insensitivity to insulin secreted by the body (Type 2 diabetes). In either case, the body is unable to efficiently move blood glucose across cell membranes to be utilized. This leads to a variety of local and systemic detrimental effects. Current treatments for diabetes focus on exogenous insulin administration and dietary control. Provided herein are treatments of diabetes using a cellular therapy to ameliorate symptoms associated with both reduced insulin secretion and insulin sensitivity. Using induced-pluripotent stem (iPS) cells, beta-like (β-like) cells similar to the endogenous insulin secreting cells were derived. These β-like cells secreted insulin in response to glucose, and corrected a hyperglycemic phenotype in a mouse model of Type 2 diabetes via an iPS cell transplant. Within the Type 2 diabetes mouse model, a long term correction of hyperglycemia was achieved as measured by blood glucose and hemoglobin Alc measurements. Reduction of hyperglycemia was also seen in a chemically-induced mouse model for Type 1 diabetes. | 12-03-2015 |
