20110001043 | DETECTOR DEVICE FOR HIGH MASS ION DETECTION, A METHOD FOR ANALYZING IONS OF HIGH MASS AND A DEVICE FOR SELECTION BETWEEN ION DETECTORS - Described here is a detector for measuring heavy mass ions with high sensitivity and low saturation for time-of-flight mass spectrometry and a detector housing for selecting between multiple detectors. It relates to sensitive measuring methods of large masses in the range of about ten thousand to a few million atomic mass units. Specifically it relates to a conversion dynode in a specifically insolated geometry followed by a discrete dynode secondary electron multiplier specifically modified to decrease electron saturation and electronic ringing. Conversion dynode detectors have been used before for time-of-flight mass spectrometry and compared to direct detection with electron multipliers they exhibit superior sensitivity for high-mass, slow-moving macromolecular ions. Using a conversion dynode specifically insolated to a common ground plane has the added capabilities of allowing an increased voltage to be applied to the conversion dynode while maintaining a minimum distance between the conversion dynode and the front of the electron multiplier. This creates faster ion flight time for the secondary ions produced within the detector allowing for higher time resolution and sensitivity from the detector. Also, by adding capacitance as charge buffers to the last few electrodes of a discrete dynode electron multiplier used as a secondary electron multiplier, saturation can be greatly reduced or avoided, which is often a major problem when measuring samples with ions covering a broad mass range. The detector housing described allows multiple detectors to be selected without breaking the vacuum. By keeping all moving mechanical parts inside the vacuum, a more simple, robust and cost effective design can be realized which provides a platform for measuring ions using different detector designs. | 01-06-2011 |

The method using High-Mass MALDI mass spectrometry provides various biological applications such as characterization of antibodies, drug discovery, and protein complex analysis including automated or higher throughput applications. | 07-21-2011 |

20110313145 | ISOLATION OF RNA - Disclosed herein are methods for purification of RNA from a sample. The RNA can be total RNA or mRNA. The method involves preparing the sample in a solution of lysis buffer and depositing into a first end of a lysis straw such that the sample solution flows through the matrix of the lysis straw and is eluted from the opposite end of the lysis straw, and depositing the eluted material into a first end of a solid phase extraction (SPE) straw, such that the deposited solution flows through the matrix of the SPE straw towards the opposite end of the SPE straw, and eluting the RNA from the SPE straw by depositing a solution of elution buffer, into the first end of the SPE straw, such that the deposited solution flows through the matrix of the SPE straw and is eluted from the opposite end of the SPE straw, wherein purified RNA from the sample is present in the eluate of the SPE straw. When the RNA is total RNA, and the sample is a cell sample, and step b) requires adding a precipitating solution to the eluted material from step a), and depositing the solution into the first end of the SPE straw, wherein the straw comprises silica microspheres, such that the deposited solution flows through the matrix of the SPE straw toward the opposite end of the SPE straw. When the RNA is mRNA and step b) further comprises depositing the solution into the first end of the solid phase extraction (SPE) straw, wherein the straw comprises oligo-dT. Pressure (e.g., at least 10 psi pressure) and heat (e.g., about 60° C.) can be applied to the samples, and/or eluates and/or straws. Examples of lysis buffers, loading buffers and elution buffers are provided. Also disclosed are the specific straws and porous polymer monolith matrix components. | 12-22-2011 |
20120077260 | RESERVOIR-BUFFERED MIXERS AND REMOTE VALVE SWITCHING FOR MICROFLUIDIC DEVICES - The present invention relates generally to the control of fluid flow rate and direction on a microfluidic device. In particular, the present invention provides an integrated valveless microfluidic device, where directional fluid control is controlled using off-chip remote valve switching and fluid flow rate changes are controlled using on-chip flow-rate changing fluid reservoirs. The present invention provides methods and systems for directional fluid control and control of fluid flow rate in an integrated microfluidic device which enables processes with different flow rates to be performed on one device without the need of on-chip valves. | 03-29-2012 |
