20100028910 | ACTIVATED PROTEIN C VARIANTS WITH NORMAL CYTOPROTECTIVE ACTIVITY BUT REDUCED ANTICOAGULANT ACTIVITY - Variants (mutants) of recombinant activated protein C (APC) or recombinant protein C (prodrug, capable of being converted to APC) that have substantial reductions in anticoagulant activity but that retain normal levels of anti-apoptotic activity are provided. Two examples of such recombinant APC mutants are KKK191-193AAA-APC and RR229/230AA-APC. APC variants and prodrugs of the invention have the desirable property of being cytoprotective (anti-apoptotic effects), while having significantly reduced risk of bleeding. The invention also provides a method of using the APC variants or prodrugs of the invention to treat subjects who will benefit from APC's cytoprotective activities that are independent of APC's anticoagulant activity. These subjects include patients at risk of damage to blood vessels or tissue in various organs caused, at least in part, by apopotosis. At risk patients include, for example, those suffering (severe) sepsis, ischemia/reperfusion injury, ischemic stroke, acute myocardial infarction, acute or chronic neurodegenerative diseases, or those undergoing organ transplantation or chemotherapy, among other conditions. Methods of screening for variants of recombinant protein C or APC that are useful in accordance with the invention are also provided. | 02-04-2010 |
20100184672 | PROTEIN C FOR USE IN MAINTAINING HEMOSTASIS - It is disclosed herein that protein C functions as a hemostatic agent. 20100284997 | DOSING REGIMEN OF ACTIVATED PROTEIN C AND VARIANTS HAVING REDUCED ANTICOAGULANT ACTIVITY - Recombinant activated protein C (APC) and APC variants with reduced anticoagulant activity were used to reduce mortality in murine models of sepsis. These models included endotoxemia and bacteremia models. We discovered that single or multiple bolus doses of APC, especially of APC variants such as RR230/231AA-APC, KKK192-194AAA-APC and 5A-APC (containing the combination of mutations present in the first two APC variants) given as a single bolus reduces 7-day mortality of mice given lethal doses of endotoxin. Administrations of a single bolus of 5A-APC after the initiation of sepsis also reduces mortality caused by LPS. 5A-APC with ≦8% of normal anticoagulant activity (which has reduced risk of bleeding) reduces mortality when given as two bolus administrations at 3 hours and then at 10 hours after initiation of bacterial infection, i.e. after onset of sepsis. This shows, first, that one or more bolus injections of APC or of APC variants, especially 5A-APC, can reduce mortality when given beginning hours after the onset of sepsis and, second, that it is not necessary to administer APC as a continuous infusion which is the current standard of practice because one or more bolus administrations can reduce mortality. Furthermore, dosages of approximately 0.06 to 0.4 mg/kg of APC and APC variants are identified to be sufficient to reduce mortality in sepsis. | 11-11-2010 |

In at least some embodiments, a medical device including a plurality of conductive elongated members that are of such a length and are connected together in such a manner to impart a degree of MRI compatibility to the device. In some embodiments, the elongated members can include a series of solid elongate members and tubular elongate members, which construction provides one way to control at least some properties of the device, for example, stiffness, torque transmission, flexibility, shape retention, and the like. In yet some additional embodiments, at least one of the elongated members has a recess into which a protrusion of another elongated member may fit. | 11-11-2010 |
20140148793 | MEDICAL DEVICE - A medical device including a shaft having an elongated inner member and an elongated tubular reinforcing member disposed over at least a portion of the inner member. In some embodiments, at least a portion of the outer surface of the inner member is spaced from the inner surface of the reinforcing member, defining a space substantially free of any other structures of the device. In some embodiments, the shaft can include a tip structure disposed on a distal portion of the inner member. In some such embodiments, the reinforcing member has a distal end, and the tip structure is disposed on the distal portion of the inner member adjacent the distal end of the reinforcing member. 20090119275 | METHOD OF MONITORING ELECTRONIC MEDIA - Consumer-generated media (CGM) and/or other media are monitored to allow an organization to become aware of, and respond to, issues that may affect how it is perceived by the public. An extract, transform, load (ETL) engine is used to process CGM and other media content, and an analytical engine utilizes a multi-step progressive filtering approach to identify those documents that are most relevant. The filtering approach includes executing broad queries to extract relevant content from different CGM and other sources, extracting text snippets from the relevant content and performing de-duplication, defining organizational identity (e.g., brand name, trade name, or company name) and hot-topic models using a rule-based and statistical-based approach, and using the models together in an orthogonal filtering approach to effectively generate alerts and reports. The methodology is found to be substantially more effective compared to a conventional keyword based approach. | 05-07-2009 |
20120179684 | SEMANTICALLY AGGREGATED INDEX IN AN INDEXER-AGNOSTIC INDEX BUILDING SYSTEM - A computer program product for an indexer-agnostic index building system includes a computer readable storage medium to store a computer readable program, wherein the computer readable program, when executed on a computer, causes the computer to perform operations for creating a semantically aggregated index. The operations include: extracting documents from a data source, wherein each document includes a data object; distributing the documents to a plurality of processing nodes within the system; for each node: indexing the data objects for each document into fields using semantic rules; and grouping indexed data objects for related fields by: classifying the documents into logical groups based on the semantic rules; and creating a searchable index shard for related logical groups. | 07-12-2012 |

20140136901 | PROACTIVE RISK ANALYSIS AND GOVERNANCE OF UPGRADE PROCESS - An incompatible software level of an information technology infrastructure component is determined by comparing collected inventory information to a minimum recommended software level. If a knowledge base search finds that the incompatible software level is associated with a prior infrastructure outage event, an outage count score is determined for the incompatible software level by applying an outage rule to a historic count of outages caused by a similar incompatible software level, and combined with an average outage severity score assigned to the incompatible software level based on a level of severity of an actual historic failure of the component within a context of the infrastructure to generate a normalized historical affinity risk score. The normalized historical affinity risk score is provided for prioritizing the correction of the incompatible software level in the context of other normalized historical risk level scores of other determined incompatible software levels. | 05-15-2014 |
