20160067314 | COMPOSITIONS AND METHODS FOR INHIBITING CHEMORESISTANCE IN CANCER AND IMPROVING RESPONSE TO THERAPY - Metastatic melanomas are highly resistant to radiation and chemotherapy from the earliest stages, which is a major factor in poor clinical outcomes. Activated leukocyte adhesion molecule (ALCAM)/CD166 was the gene that showed the highest correlation with detachment-induced chemoresistance. SiRNA-mediated depletion or antibody blocking of ALCAM specifically inhibited the increase in chemoresistance after detachment. This antibody also improved chemotherapeutic responses in a mouse xenograft model of human melanoma. Previous studies identified ALCAM as a marker for tumor aggressiveness and poor prognosis, and as a marker for “stemness”. Targeting ALCAM may therefore represent a novel approach for treatment of otherwise intractable melanomas. It was also found that stimulating integrin signaling enhanced chemosensitivity of melanoma to chemotherapeutic agents. The present invention provides a novel multimeric peptide construct comprising fibronectin fragments useful for stimulating integrin signaling and for enhancing chemosensitivity of melanomas. | 03-10-2016 |

20150370944 | SYSTEM FOR PLACEMENT OPTIMIZATION OF CHIP DESIGN FOR TRANSIENT NOISE CONTROL AND RELATED METHODS THEREOF - Transient voltage noise, including resistive and reactive noise, causes timing errors at runtime. A heuristic framework, Walking Pads, is introduced to minimize transient voltage violations by optimizing power supply pad placement. It is shown that the steady-state optimal design point differs from the transient optimum, and further noise reduction can be achieved with transient optimization. The methodology significantly reduces voltage violations by balancing the average transient voltage noise of the four branches at each pad site. When pad placement is optimized using a representative stressmark, voltage violations are reduced 46-80% across 11 Parsec benchmarks with respect to the results from IR-drop-optimized pad placement. It is shown that the allocation of on-chip decoupling capacitance significantly influences the optimal locations of pads. | 12-24-2015 |

20150297680 | COMPOSITIONS AND METHODS FOR INCREASING PROTEIN HALF-LIFE - In order to extend the serum half-life of a protein, we exploited the site-specific fatty acid-conjugation to a permissive site of a protein, using copper-catalyzed alkyne-azide cycloaddition, by linking a fatty acid derivative to p-ethynylphenylalanine incorporated into a protein using an engineered pair of yeast tRNA/aminoacyl tRNA synthetase. As a proof-of-concept, we show that single palmitic acid conjugated to superfolder green fluorescent protein (sfGFP) in a site-specific manner enhanced a protein's albumin-binding in vitro about 20 times and the serum half-life in vivo 5 times when compared to those of the unmodified sfGFP. Furthermore, the fatty acid conjugation did not cause a significant reduction in the fluorescence of sfGFP. Therefore, these results clearly indicate that the site-specific fatty acid-conjugation is a very promising strategy to prolong protein serum half-life in vivo without compromising its folded structure and activity. | 10-22-2015 |

20140364523 | Viscoelastic Silicone Rubber Compositions - This invention relates to viscoelastic silicone rubber compositions that are the products of the reaction of: (a) a silanol-terminated polyorganosiloxane base; (b) a boron-containing crosslinking agent; and (c) a siloxane bond-forming crosslinking agent. In a viscoelastic silicone rubber composition of the invention some of the crosslinks, the siloxane crosslinks, are permanent and others of the crosslinks, the boron-containing crosslinks, are temporary. Because a fraction of its crosslinks can come apart and then reform, a viscoelastic silicone rubber composition of the invention can relax stress in response to strain and thus adapt to new shapes. The composition has sufficient permanent crosslinks, however, to establish a permanent equilibrium shape to which the composition will eventually return when not subject to any imposed stress. A viscoelastic silicone rubber composition has sufficient temporary crosslinks to give the composition a stiffness that is greater on short timescales than it is on longer timescales. | 12-11-2014 |

20120285114 | Manufacture of Lattice Truss Structures from Monolithic Materials - Methods and systems to manufacture lattice-based sandwich structures from monolithic material eliminate the bonding process which is conventionally used to join lattice-based truss cores to facesheets to form sandwich structures. The bonded interface is a key mode of failure for sandwich structures which are subjected to shear or bending loads because the nodes transfer forces from the facesheets to the core members while the topology for a given core relative density dictates the load carrying capacity (assuming adequate node-bond strength exists). In one aspect, a core and related structures provide very low density, good crush resistance and high in-plane shear resistance. Another aspect of the truss structures may include sandwich panel cores and lattice truss topology that may be designed to efficiently support panel bending loads while maintaining an open topology that facilitates multifunctional applications. | 11-15-2012 |

20120252031 | COMPOSITIONS AND METHODS FOR REGULATING SAS1R - The present invention provides compositions and methods useful for regulating fertilization and for use as a contraceptive based on the discovery herein of an oocyte specific protein, SAS1R (Sperm Acrosomal SLLP1 Receptor), which is a sperm protein receptor. Six SAS1R variants, including the full length SAS1R, were identified. mSLLP1 and SAS1R co-localized to oocytes and to acrosomes of acrosome-reacted sperm. Interactions between mSLLP1 and SAS1R were demonstrated by far-western analysis, in a yeast two-hybrid system under stringent selection conditions, and by immunoprecipitation of SAS1R by anti-mSLLP1 as well as the converse. Purified recombinant SAS1R was found to have protease activity, to inhibit fertilization in-vitro, and to induce an immune response in females. Together, the results suggest SAS1R is a proteolytically active, oocyte and early embryo specific oolemmal metalloprotease receptor for the sperm intra-acrosomal ligand SLLP1 and is a target for regulating fertilization and as a contraceptive. | 10-04-2012 |

Many materials (metals, ceramics, glasses, polymers, composites and even semiconductors) can be shaped into cellular, truss-like architectures with open, closed or mixed types of porosity and then very uniformly arranged in controlled, three-dimensional space-filling arrays. The truss-like elements do not necessarily have a constant cross-section, nor are they necessarily straight or solid throughout (they could be hollow). Their cross sections can be circular, square, triangular, I-beam or other shapes of interest depending on multifunctional needs. When bonded together by solid state, liquid phase, pressing or other methods at points of contact, a cellular structure of highly repeatable cell geometry and few imperfections results. The bonds hold the truss elements together in a desired configuration, allow load to be efficiently transferred amongst them and make the resulting structure significantly more rigid when bent, compressed or sheared. These constructed cellular solids offer a broad range of multifunctional structural uses with a tremendous freedom for choosing the truss type, orientation and distribution. Multiple materials can be intermixed. | 10-13-2011 |
