Role of IRS-2 in insulin and cytokine signalling

Article Abstract:

The IRS-2 signalling protein, purified and cloned from myeloid progenitor cells, are essential for signalling by many receptor systems. The identification of IRS-2 gives new insight into the modular structure and function of IRS signalling proteins. IRS-2 resembles IRS-1 in many respects. Both of these IRS signalling systems help in signal amplification by removing the stoichiometric constraints in receptors that provide SH2 proteins to their autophosphorylation sites.

Disruption of IRS-2 causes type 2 diabetes in mice

Article Abstract:

Disruption of insulin-receptor substrates (IRS proteins) in mice retards growth, although diabetes does not develop as insulin secretion increases to compensate. Disruption of IRS-2 is shown to impair both peripheral insulin signalling and pancreatic Beta-cell function, and IRS-2-deficient mice exhibited progressive deterioration of glucose homeostasis. It is suggested that IRS-2 dysfunction may contribute to the pathophysiology of human type 2 diabetes.

Article Abstract:

Research is presented concerning the influence of dietary restrictions and obesity on the fertility of mammals. The depletion of insulin receptor substrate-2 which also causes infertility is discussed.
