Alzheimer-type neuropathology in transgenic mice overexpressing V717F beta-amyloid precursor protein
Article Abstract:
Transgenic mice that overexpress human mutant beta-amyloid precursor protein exhibit symptoms of Alzheimer's disease (AD) such as synaptic loss, neuritic plaques, astrocytosis and microgliosis. In this mutant protein, a phenylalanine replaces valine in the 717 residue. These data suggest a role for such transgenic mice in testing drugs for AD treatment.

Immunization with amyloid-beta attenuates Alzheimer-disease-like pathology in the PDAPP mouse
Article Abstract:
Immunization with A-beta-42, the 42-amino-acid form of amyloid-beta peptide, prevents the development of Alzheimer's disease-like neuropathology in PDAPP transgenic mice, which overexpress mutant human amyloid precursor protein. In these mice, immunization with A-beta-42 disrupts a key mechanism of amyloid plaque formation. Immunization either prevents deposition and/or boosts the clearance of A-beta from the brain. It is likely that a similar reduction of neuropathology in Alzheimer's disease patients would be clinically beneficial.
