A quantitative protein interaction network for the ErbB receptors using protein micro arrays

Article Abstract:

The protein micro arrays used are virtually comprising every Src homology 2 (SH2) and phosphotyrosine binding (PTB) domain encoded in human genome to measure the equilibrium dissociation constant of each domain for 61 peptides representing physiological sites of tyrosine and phosphorylation on the four ErbB receptors. This involved 77,592 independent biochemical measurements and provided a quantitative protein interaction network that reveals many new interactions.

Article Abstract:

The functional importance of phosphorylation-dependent modulation of transcription factor activity is discussed. This evidence of a nuclear transcriptional coactivator with intrinsic phosphate activity suggests an unanticipated method of fine-tuning transcriptional regulation.

The Eyes absent is also a protein tyrosine phosphates is demonstrated. The eyes absent proteins are members of a conserved regulatory network implicated in the development of the eye, muscle, kidney and ear.
