A genetic link between co-suppression and RNA interference in C. elegans

Article Abstract:

Mutants of Caenorhabditis elegans that are defective in transposon silencing and RNA interference are also resistant to co-suppression. This research showed co-suppression phenotypes for only those mutants that affect RNA interference and transposon silencing together. These mutants appear to affect both DNA and RNA. These findings were in alignment with a model of transposon detection through double-stranded RNA, as transcription of transgenes appears to be a prerequisite for co-suppression.

A micrococcal nuclease homologue in RNai effector complexes

Article Abstract:

Experimentally it is shown that Tudor-SN (tudor staphylococcal nuclease) is a component of the RISC (RNA-induced silencing complex) enzyme in Caenorhabditis elegans, Drosophila and mammals. The molecular structure and nuclease activity of Tudor-SN are explained.

Processing of primary microRNAs by the microprocessor complex

Article Abstract:

It is shown that Drosha exists in a multi protein complex, the microprocessor, and begin the process of deconstructing that complex into its constituent components. Suppression of Pasha expression in Drosophila cells or Caenorhabditis elegans interferes with pri-miRNA processing, leading to an accumulation of pri-miRNAs and a reduction in mature miRNAs.
