A novel series of P2T purinoceptor antagonists: definition of the role of ADP in arterial thrombosis

Article Abstract:

The advantages of aspirin as an antithrombotic agent are economics and its relatively safe mode of administration. However, aspirin has limited clinical efficacy and can only interfere mechanistically with thromboxane-induced platelet aggregation. There is a new class of agents which antagonize the aggregatory actions of adenosine diphosphate at the P2T purinoceptor on the platelet membrane. Unlike adenosine triphosphate, these new agents are potent, selective P2T purinoceptor antagonists both in vitro and in vivo.

Deviations from receptor theory?

Article Abstract:

A study was conducted to determine the different response levels of agonists using three muscarinic acetylcholine receptor agonists on adenylyl cyclase activity in the rat corpus striatum and in rabbit myocardial membranes. Results showed a curve data which failed to match the model curve. However, various methods of analyses showed that the data can fit the model if no restrictions are used. This proves that the traditional receptor model can accomodate varying data sets.

An insight into arterial thrombosis?

Article Abstract:

Researchers have developed a non-activating monoclonal antibody to the non-integrin GPVI, a transmembrane protein believed to be involved in the activation of platelets by collagen. This could lead to a treatment for arterial thrombosis.
