Interaction of carnitine and propionate with pyruvate oxidation by hepatocytes from clofibrate-treated rats: importance of Coenzyme A availability

Article Abstract:

Propionate disrupts normal liver function after the accumulation of acylcoenzyme As. Clofibrate can regulate the toxic effects of propionate in the liver by increasing the total amount of coenzyme A. Hepatocytes from clofibrate-treated rats were isolated and used to study its effect on the interactions of propionate and carnitine with pyruvate oxidation. Propionate exhibited a decreased inhibitory effect. The antitoxic effects of carnitine did not increase because carnitine also induced a decrease in total coenzyme A content.

L-carnitine administration and withdrawal affect plasma and hepatic carnitine concentrations, plasma lipid and lipoprotein composition, and in vitro hepatic lipogenesis from labeled mevalonate and oleate in normal rabbits

Article Abstract:

The effects of L-carnitine administration and withdrawal on lipid and lipoprotein metabolism were investigated. The results showed that carnitine administration promotes increases in plasma and hepatic carnitine levels. However, upon withdrawal, only the plasma levels returned rapidly to normal. In addition, withdrawal of carnitine led to reductions in plasma total cholesterol and VLDL cholesterol, and increases in IDL, LDL and HDL proteins. Moreover, carnitine withdrawal also altered hepatic lipid synthesis profiles.
