Article Abstract:
A procedure was developed for immobilization of Listeria monocytogenes cells on metal hydroxides coupled with detection and enumeration using an automated optical system. The results of the immobilization procedure (< 1 h) and detection during overnight incubation agreed with calculated plate counts, and this technique is simple and rapid and provides samples that are ready for confirmation of the presence of the pathogen by rapid methods.

Microbiological research using the pulsed-field technique of contour-clamped-homogeneous-electric-field electrophoresis has produced a circular map of the facultative intracellular pathogen Listeria monocytogenes Scott A serotype 4b strain. The temperature-sensitive vectors pLUCH80 and pLUCH88 were constructed prior to map formation using a Tn917 derivative (Tn917::NotI/SmaI) containing NotI and SmaI sites.

The effects of nine common food industry stresses on the times to the turbidity distribution of Listeria monocytogenes are determined. According to the studies performed, results suggest that stress-induced variability might be important in determining the reliability of predictive microbiological models.
