Article Abstract:
Dichloromuconate is the inducing agent of the 2,4-dichlorophenoxyacetic acid (2,4-D) pathway of Alcaligenes eutrophus JMP134 (pJP4) plasmid. The identification is based on the analysis of beta-galactosidase gene lacZ transcriptional fusions with tfd structural genes in the presence and absence of 2,4-D. Fusions of individual genes help in reporter and disrupter activities of gene expression. Increased reporter activities are evident in fusions occurring in genes that encode enzymes, which function after the production of the inducing intermediate.

Researchers designed a plasmid that can increase the mutation frequency of a bacterium by 20- to 4,000-fold. Once the desired trait is achieved, the plasmid can be removed, returning the bacterium to its normal mutation rate. This technique could be used to develop bacteria that can biodegrade high concentrations of metabolites and solvents.

A replicon from the lactococcal plasmid pCD4 was used to introduce a mechanism for phage resistance into Lactococcus lactis strain MG1363 and two other industrial strains. The phenotype lasted over 100 generations and provided resistance to phage species 936 and c2.
