Article Abstract:
Pseudomonas putida KT2440 rpoH gene regulation is discussed based on molecular characterization. The gene encodes the heat shock sigma factor sigma (super.32). An inverted repeat sequence 20 bp downstream of the rpoH stop codon acts as a terminator in vivo in P. putida growing at 18 degrees C to 42 degrees C.

Results show that two DNA segments from Sinorhizobium meliloti exhibit sequence similarity to rpoH genes, encoding the sigma(sup)32 RpoH heat -shock transcription factor. Of the two genes, rpoH1 and rpoH2, the former upon mutation impairs growth and affect nitrogen fixation during symbiosis with alfalfa.

Article Abstract:
The solvent tolerance of three toluene-degrading Pseudomonas putida strains and Pseudomonas mendocina and their survival in soil after solvent shock are analyzed. The study revealed that the viability of P. putida DOT-T1 and F1 did not disappear upon heptane and propylbenzene shock. It was severely affected, however, by toluene shock even if both strains recovered and colonized polluted niches. This recovery was dependent upon soil humidity and the incubation temperature.
