Identification of procaryotic repetitive DNA suitable for use as fingerprinting probes

Article Abstract:

A method which enables the rapid identification of procaryotic repetitive sequences for use as DNA fingerprinting probes was developed. The technique involves shotgun cloning of restricted genomic DNA and selection of clones containing repetitive DNA. Confirmation that the clones contained repeated sequences was by Southern hybridization. The sequences were used for specific and sensitive detection of bacteria and as target sequences for integration of reporter genes.

A computer analysis of primer and probe hybridization potential with bacterial small-subunit rRNA sequences

Article Abstract:

A new bacterial identification protocol involving fluorescent labeling and polymerase chain reaction has been developed. The protocol, which simulates hybridization between DNA sequences, was used to examine the SSU rRNA sequences in the Ribosomal Database Project bank. Analysis suggested that length differences in terminal restriction fragments of polymerase chain reaction products from the SSU rRNA sequences can pinpoint several bacteria.

Differences in the hybridization pattern of Bacillus subtilis genes coding for rRNA depend on the method of DNA preparation

Article Abstract:

DNA was prepared from Bacillus subtilis by organic deproteinization, potassium acetate deproteinization and the utilization of cetyltrimethylammonium bromide. Hybridization with an rDNA probe done afterwards reveled conflicting patterns of gene coding. Results indicate that the presence of peptidoglycan in the purified B. subtilis strain restricted access to the DNA.
