Identification of four phage resistance plasmids from Lactococcus lactis subsp. cremoris HO2

Article Abstract:

Novel latococcal bacteriophage resistance plasmids from 16 strains of Lactococcus lactis subsp cremoris HO2, which originated from a mixed strain Cheddar cheese starter culture, were identified and characterized. Six endogenous plasmids were identified, four of which were found to either confer or affect bacteriophage resistance mechanisms of different strengths and specificities. pCI658 was associated with adsorption inhibition, R/M was expressed through the combined presence of pCI642 and pCI605 and the plasmic pCI646 exerts phage resistance to some extent.

Controlled integration into the Lactococcus chromosome of the pCI829-encoded abortive infection gene from Lactococcus lactis subsp. lactis UC811

Article Abstract:

The abi gene codes for an abortive infection defense mechanism which was originally identified in Lactococcus lactis subsp. lactis UC811. The abi sequence was integrated into the L. lactis subsp. lactis CH919 chromosome using the integration plasmid pCI194. Chromosomal integration resulted in a phage insensitivity phenotype which was less intense than that observed for the plasmid-borne gene. However, increased expression of phage insensitivity could be achieved by amplifying the abi sequence.

ScrFI restriction-modification system of Lactococcus lactis subsp. cremoris UC503: cloning and characterization of two ScrFI methylase genes

Article Abstract:

The cloning and expression of two methylase genes in Escherichia coli is discussed. The linkage of the homology-absent genes to the Lactococcus genome was established by the inverse polymerase chain reaction of flanking chromosonal DNA. Sequencing of the DNA and amino acids was also conducted. The importance of DNA-protein interactions in most biological processes was demonstrated.
