Article Abstract:
A Bifidobacterium genus-specific target sequence in the V9 variable region of the 16S rRNA was analyzed and utilized to develop a hybridization probe. All of the 30 type strains of Bifidobacterium which are available at the German culture collection Deutsche Sammlung von Mikroorganismen und Zellkulturen, six commercially available production strains, and 34 closely related relevant strains were tested. All tested bifidobacteria exhibited distinct positive signals by colony hybridization, whereas all negative controls had no distinct dots except Gardnerella vaginalis DSM4944 and Propionibacterium freudenreichii subsp. shermanii DSM4902 which had slight signals.

Article Abstract:
Hybridization probes targetting 16S rRNA sequences appear to allow genus- specific detection of bifidobacteria in human fecal samples. Fluorescent probes based on the V2, V4 and V8 variable regions of 16S rRNA unique to the bacterium were used for quantitative in situ hybridization in samples on glass slides. The technique revealed that culture techniques may be overestimating the bifidobacteria population size in relation to total intestinal microflora.

Article Abstract:
16S rRNA gene probes were generated to monitor the growth of Bifidobacterium sp. strain LW420 in infant fecal samples. Sequencing and analysis of DNA from single-strain and mixed cultures of bifidobacteria and from heterogeneous feces samples revealed a significant homology of 16S rDNA of LW420 with known 16S rDNA sequences. PCR analyses showed that all fecal samples was positive for Bifidobacterium.
