Article Abstract:
A study was conducted to determine the influence of peroxidase production and enzyme profiles on xenobiotic compound oxidation. Effects on a polycyclic aromatic hydrocarbon anthracene compound and the ligninolytic indicator dye Poly R-478 were analyzed. The combined presence of manganese and veratryl alcohol was found to have a synergistic negative effect on the oxidation of anthracene and the decolorization of Poly R-478. Results indicated that increased H2O2-generating oxidase production could be achieved by improving the physiology or strain of white rot fungi for the degradation of xenobiotic compounds.

The crude laccase from Trametes versicolor oxidizes anthracene and the carcinogen benzo(a)pyrene, polycyclic aromatic hydrocarbons, which have ionization potentials of less than or equal to 7.45 eV. The cooxidant 2,2'-azinobis(3-ethylbenzthiazoline-6-sulfonate) (ABTS) increases benzo(a)pyrene oxidation. ABTS is necessary for the oxidation of benzo(a)pyrene by purified laccase isozymes. Although ABTS is unnecessary for anthracene oxidation by purified laccase isozymes, it enhances the oxidation. Anthracene oxidation increases in the presence of low-molecular-weight culture fluid ultrafiltrate.

Article Abstract:
The biobleaching of oxygen-delignified eucalyptus kraft pulp by Bjerkandera sp. strain BOS55 was analyzed to determine the role of manganese-dependent peroxidase (MnP) on the biosynthetic activity of the phytopathogenic fungi. Supplementation of Bjerkandera sp. strain BOS55 cultures with standard basal mineral nutrients containing manganese did not affect the delignification and bleaching eucalyptus kraft pulp. Furthermore, MnP was the predominant enzyme produced by Bjerkandera sp. strain BOS55 during the bioleaching of oxygen delignified kraft pulp.
