Article Abstract:
A study was conducted to describe two mammalian RNA-binding Tra2 proteins. It was observed that human Tra2 proteins are present in HeLa cell nuclear extracts and that they bind efficiently and specifically to a characterized pre-messenger RNA splicing enhancer element. Both Tra2 proteins activated enhancer-dependent splicing in a sequence-specific manner and restored after inhibition with competitor RNA. These findings indicate that mammalian Tra2 proteins are sequence-specific splicing activators that likely participate in the control of cell-specific splicing patterns.

Article Abstract:
An overview of earlier studies as well as recent findings that support the existence of an evolutionarily conserved role for cotranscriptional processes in the maintenance of genome stability is presented. The way by which different species protect the integrity of their genomes from potentially disastrous effects of nascent transcripts by diverse mechanisms and the possible mechanisms by which if unchecked, nascent transcripts can lead to genome instability is discussed.

The importance of alternative splicing factor/splicing factor 2 (ASF/SF2) for maintenance of genomic stability is discussed. Recruiting ASF/SF2 to nascent transcripts by RNA polymerase II prevents the formation of mutagenic R loop structures.
