Hedgehog/Ras interactions regulate early stages of pancreatic cancer

Article Abstract:

The generation of a novel mouse model of pancreatic cancer based on epithelium-specific activation of the Hedgehog signaling pathway is described. The results have indicated the cooperation of Hedgehog and Ras signaling during the earliest stages of pancreatic ductal adenocarcinoma (PDA) formation and they have also marked Hedgehog pathway components as relevant therapeutic targets for both early and advanced stages of pancreatic ductal neoplasia.

Persistent expression of PDX-1 in the pancreas causes acinar-to-ductal metaplasia through Stat3 activation

Article Abstract:

The transcription factor pancreatic and duodenal homeobox factor 1 (PDX-1) was constitutively expressed in all pancreatic lineages by transgenic approaches in order to determine whether sustained expression of PDX-1 could affect pancreas development. The results provide a mechanism of pancreatic metaplasia by which persistent PDX-1 expression cell-autonomously induces acinar-to-ductal transition through Stat3 activation.

Aggressive pancreatic ductal adenocarcinoma in mice caused by pancreas-specific blockade of transforming growth factor-[beta] signaling in cooperation with active Kras expression

Article Abstract:

The effect of type II transforming growth factor-[beta] receptor (Tgfbr2) knockout on pancreas development and further on pancreatic ductal adenocarcinoma (PDAC) progression in the context of active Kras expression is examined. Results reveal that blockade of TGF-[beta] signaling and activated Ras signaling cooperate to promote PDAC progression.
