Article Abstract:
Genetic experiments enable the investigation of the type of germinal transpositions of artificial dissociation (Ds) transposons, and their behavior and characteristics in tomato. A marker gene displaces the activator transposae gene to produce the Ds element. Examination of germinal excision frequencies reveals the large frequency variation between and within plants, and the frequencies of Ds averaged 15-40%. The extent of clonal transposition events among sibling offspring are determined using Southern hybridization analysis.

Article Abstract:
The molecular basis of maize Activator (Ac) transposition in relation to DNA replication was elucidated. Southern analyses were conducted on Ac donor and target sites before and after insertion. The analyses showed that Ac is excised from only one of the daughter chromosomes after DNA replication. Insertion of Ac can occur in both replicated and unreplicated target sites. In addition, most of the transposition events occur during the period of DNA synthesis or shortly thereafter.

Article Abstract:
Ac reinsertion along the length of the P gene in maize after transposition of the element into adjacent chromosomal regions produced more than 250 unstable alleles for variegation. The variegation patterns are indicative of the location of the Ac reinsertion which can be anywhere within the 12.3 kilobase segment spanning the promoter, exon and intron regions of the P gene. This demonstrates that Ac can translocate to adjacent sites flanking the donor region.
