Article Abstract:
Deletion mutagenesis is used for fine genetic mapping of mutations and DNA markets in several plant species. Gamma-irradiated haploid wild-type pollen was used to pollinate diploid and tetraploid multimarker lines of the model higher plant Arabidopsis. Chromosome loss mutations and large deletions were found to be induced in the irradiated pollen, but were eliminated in diploid M1 plants and could be rescued in triploid M1 progeny.

Article Abstract:
Nonchromosomal strip (NCS) mutants of maize carry deletions of some essential mitochondrial genes. NCS deletion mutations appear to result from rare recombination over small repeated sequences. It is reported that homoplasmic nad4 deletion mutant cultures can be easily generating and maintained from ears of NCS2 striped plants. Also the first case of a rare hompoplasmic nad4 deletion mutant plant is reported.

Article Abstract:
The presence of mitochondrial heteroplasmy is demonstrated in the cytochrome oxidase 1 (CO1) gene of the brittle star Astrobrachion constrictum. Distinct single-strand conformation polymorphism (SSCP) haplotypes differing by two substitutions were found, and there was evidence for heteroplasmy. The haplotypes were isolated and characterized using polymerase chain reaction cloning, SSCP and sequencing.
