Genetic diversity of rhizobial symbionts isolated from legume species within the genera Astragalus, Oxytropis, and Onobrychis

Article Abstract:

A research was conducted to investigate the genetic diversity of rhizobial isolates from Astragalus, Oxytropis and Onobrychis spp. originating from several geographic sites using mapped restriction site polymorphism analysis of 16S rRNA genes and by a polymerase chain reaction DNA fingerprinting technique. Each strain was assigned a 16s rDNA type defined by restriction patterns obtained with restriction endonucleases. Results indicated that rhizobial classification was independent of origin and that there were nine distinct 16S rDNA types among the 35 strains with the genus Mesorhizobium.

Typing of rhizobia by PCR DNA fingerprinting and PCR-restriction fragment length polymorphism analysis of chromosomal and symbiotic gene regions: application to Rhizobium leguminosarum and its different biovars

Article Abstract:

A PCR-DNA fingerprinting technique is effective in typing strains of rhizobia. The method gives the same resolution as conventional restriction fragment length polymorphism (RFLP) techniques. The technique is quick and is useful for the detection and study of large populations of bacteria at the intraspecies level. The classification of strains has no dependence on the biovar status. PCR-RFLP analysis of nifDK and nodD gene regions was used to detect intrabiovar variations.

Article Abstract:

Results points out that lateral genetic transfer of nodC and nifH genes across rhizobial species lead to diversification and structuring in rhizobial populations. Genetic analysis reveals a close relationship of rhizobial genes with those of Phaseolus vulgaris symbionts' genes.
