Geminin, an inhibitor of DNA replication is degraded during mitosis

Article Abstract:

Synchronized cultured HeLa cells were used in the investigation of the metabolism of geminin during the cell cycle. Results reveal that geminin is starts accumulating at the S phase of the cell cycle and during the G2 and M phases. It is degraded during the transition from metaphase to anaphase. Geminin inhibits DNA replication during the S, G2 and M phases by preventing the MCM complex incorporation into the prereplication complex.

The molecular basis for phosphodependent substrate targeting and regulation of PIks by the Polo-box domain

Article Abstract:

Results reveal that Polo-box domain in Polo-like kinases recognize phosphoserine/threonine containing motiffs and the phosphopeptides bind to a positively charged cleft in the Polo-box interface. Data indicate that phosphopeptide binding to Polo-box domain stimulates kinase activity in Polo-like kinases, which undergo conformational switching for regulatory function.

Tome-1, a trigger of mitotic entry, is degraded during G1 via the APC

Article Abstract:

Results reveal that the cytosolic protein Tome-1 is essential for activation of relevant enzymes to ensure mitotic entry. Data suggest that Tome-1 in combination with Skp-1 degrades the kinase wee 1 thereby facilitating mitotic entry in Xenopus egg extracts.
