First evaluation of nuclear DNA content in Setaria genus by flow cytometry

Article Abstract:

Flow cytometry was utilized to obtain nuclear DNA content, base composition and ploidy levels in several Setaria species which includes the domesticated plant foxtail millet. Root tips were treated with 8-hydroxyquinolin at 16 degrees C while descriptors from various nomenclatures were used to determine botanical identification. Results indicated the conservation of cereal genomes based on the colinearity of the species' maps.

rDNA organization and evolutionary relationships in the genus Hypochaeris (Asteraceae)

Article Abstract:

The pattern of rDNA sequence distribution in 11 Hypochaeris species from South America and Europe was observed by in situ hybridization utilizing probes of 18S-5.8S-28S and 5S ribosomal RNA genes. The rDNA localization revealed that chromosomes rearrangements as centric shifts, centric fusions and inactivation of rDNA genes happened during the course of evolution of Hypocharies species from the two geographical areas.

Evolutionary DNA variation in the genus Hypochaeris

Article Abstract:

The pattern of nuclear DNA variations in 8 Hypochaeris species was established by flow cytometry. Phylogenetic analysis suggests the high probability that genome size evolved by losing DNA content in some lineages and by gain in another. No correlation between 2C DNA content and GC% was discovered, so neither GC nor AT fractions were preferentially associated with size in genome.
