Article Abstract:
Research was conducted to examine the expression of the isoamylase gene of Flavobacterium odoratum KU in Escherichia coli and to identify essential residues of the enzyme by site-directed mutagenesis. The objective is to gain an insight into the relationship between the function and structure of isoamylase by sequencing the entire isolated DNA fragment and deducing the primary sequence of isoamylase of F odoratum. Results indicate that the bacterial and plat isoamylase but not the glycogen operon protein had a longer fourth loop than the other amylases.

The cloning of a novel aldo-keto reductase gene from Klebsiella sp. strain F51-1-2 and its functional expression in Escherichia coli was studied. Assays confirmed that reductive transformation of organophosphorous compounds by the recombinant AKR5F1 specifically required NADH.
