Article Abstract:
A study was conducted of drug resistance induction through intrachromosomal mammalian gene amplification. The triggering of breakage-fusion-bridge cycles was correlated to induction of fragile site expression. The findings suggest that site-specific breaks triggered by fragile site inducers are responsible for the early features of intrachromosomal amplification. Fragile sites play a very important role in the amplification of at least some oncogenes during tumor progression.

Article Abstract:
Research indicates that DNA replication initiation sites exhibit AT richness and oriGNAI3 is a dominat origin of the domain that establishes large interorigin distances along chromosomes in mammalian somatic cells. Data show that the efficiency of initiation at oriGNAI3 is decreased with the activation of other dormant origins, suggesting that nucleotide pools are involved in the assignment of active origins.

Article Abstract:
The role of palindromic sequences in gene amplification is examined by analyzing budding yeasts. The disruption of DNA damage and DNA replication checkpoints in yeast has led to an increase in the frequency of the unusual rearrangements prompted by instability at fragile sites just like in mammalian cells.
