Article Abstract:
The expression cloning and analysis of murine and human 2-5A-dependent RNAases is reported. 2-5A-dependent RNAase is an interferon-induced enzyme which is activated by 5'-phosphorylated, 2',5'-linked oligoadenylates (2-5A). It is implicated in the molecular mechanisms of interferon action and the control of RNA stability in mammalian cells. The results showed similarity of 2-5A-dependent RNAase to RNAase E, an enzyme involved in mRNA stability in Escherichia coli. Deletion analysis and site-directed mutagenesis showed that a duplicated phosphate-binding loop motif is involved in 2-5A binding.

Article Abstract:
The interaction between odorants and olfactory receptors was examined using an olfactory receptor library expression. A total of 80 chimeric receptors from this library were evaluated against 26 odorants after being transfected into HEK-293 cells. The analysis identified three receptors that responded to micromolecular concentrations of carvone, (-)citronellal and limonene. The study also revealed that the mouse 17 receptor favors heptanal instead ofoctanal due to a single valine-to-isoleucine substitution.

Article Abstract:
A novel member of the otx homeodomain gene family, Crx, was isolated and characterized through reporter constructs for a transactivation assay of photoreceptor cells in the neural retina. Results revealed that CRX exhibited specific binding to conserved DNA sequences found upstream of several photoreceptor-specific genes. CRX was able to activate transcription from the minimum promoter in a CRX-binding site-dependent manner.
