Evaluation of molecular typing techniques to assign genetic diversity among Saccharomyces cerevisiae strains

Article Abstract:

An evaluation of the resolution of the polymerase chain reaction (PCR)-mediated typing techniques indicates that no single technique is sufficient for the discrimination of Saccharomyces cerevisiae strains. The random amplified polymorphic assay using the OPA-11 and 24 primers and PCR fingerprinting allow for the study of genetic diversity between the strains. Restriction enzyme analysis of PCR-amplified units is useful in determining the intraspecies-specific patterns.

Methods for integrated air sampling and DNA analysis for detection of airborne fungal spores

Article Abstract:

Nested polymerase chain reaction (PCR) is better for detecting fungal spores in air samples than single-step PCR or PCR followed by Southern blotting. Disrupting the spores before PCR analysis was also more effective in maximizing counts than using untreated spores.

The MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay is a fast and reliable method for colorimetric determination of fungal cell densities

Article Abstract:

A method is described for estimating the number of viable fungal cells in small samples of liquid culture. It uses a colorimetric method and is faster and more accurate than hemocytometer counting.
