Article Abstract:
The correlation between the indirect fluorescent antibody (IFA) staining method and PCR detection is 100% for Giardia cysts and just a little lower for Cryptosporidium. A double PCR is used for Giardia and a nested PCR method is used to detect Cryptosporidium. Treatment processes produce a 3-log reduction of Giardia cysts and a 2-log reduction of Cryptosporidium oocysts. A vortex flow filtration (VFF) technique is used for concentration before the recommended American Society for Testing and Materials method for processing by indirect fluorescent antibody staining. The VFF is also useful for processing drinking water.

Article Abstract:
The Colilert-Marine Water (MW) product was subjected to a premarketing evaluation. This involved parallel testing with the multiple tube fermentation method for 86 coastal water samples. Paired t tests and linear regression were performed as the bacterial isolates were assessed by genetic and biochemical analysis. The utility of the Colilert MW method for monitoring coastal beach waters was demonstrated.

A technique for detecting the presence of potentially pathogenic microorganisms in sewage and sludge was developed. This involved the utilization of a rapid purification procedure and a rapid extraction method combined with polymerase chain reaction. The method was successfully applied for the amplification of the Escherichia coli uidA gene from both sewage and sludge fragment.
