Article Abstract:
Propidium monoazide (PMA) and ethidium monoazide are used for enumeration of viable Listeria monocytogenes cells in the presence of dead cells. PMA does not have any antimicrobial effect on Listeria monocytogenes and viable cell counts are related to real-time PCR threshold cycle values for PMA-treated cells from plaktonic and biofilm sources.

Four experiments performed to study the usefulness of propidium monoazide (PMA) treatment of mixed bacterial communities comprising intact and compromised cells in combination with end-point PCR. The two experiments suggest that PMA treatment affects banding patterns in denaturing gradient gel electrophoresis community profiles.

A new concept is developed with the viable/dead stain ethidium monoazide (EMA) in combination with real-timePCR (EMA-PCR). EMA-PCR offers a novel real-time PCR method for quantitative distinction between viable and dead cells with potentially very wide application.
