Article Abstract:
A flow cytometry method utilizing the nucleic acid stain SYBR Green I is effective for cell enumeration and cell cycle analysis of natural populations of picoplankton in oceanic environments. SYBR Green I distinguishes heterotrophic bacteria and autotrophic Prochlorococcus cells, unlike the TOTO-1 and TO-PRO-1 stains. It also distinguishes the two groups of heterotrophic bacteria. The DNA distribution resolution obtained with SYBR Green I is the same as that obtained with Hoechst 33342. It also allows the analysis of photosynthetic prokaryotes over the whole water column.

Article Abstract:
Flow cytometric analyses are conducted for cultures of Escherichia coli and a variety of marine prokaryotes using novel blue light-excited fluorescent dyes for nucleic acid. Results suggest that the dyes are very sensitive to ionic strength, making them unsuitable for cell cycle analysis in natural samples. However, the dyes are very useful for enumerating prokaryotic cells by flow cytometry.

Light appears to trigger DNA replication and cell division in Prochlorococcus strains. In total darkness, cells were arrested in the G(sub 1) phase and in the S phase.
