Enhancer regions responsible for temporal and cell-type-specific expression of a spore coat gene in Dictyostelium

Article Abstract:

Portions of the upstream region of the cotB gene in Dictyostelium discoideum was fused to lacZ to enable beta-galactosidase to accumulate and mark prespore cells. A random distribution was observed among prespore cells throughout the aggregates, but these cells eventually separated from pre-stalk cells. Two distinct regions which provide proper temporal and cell-type regulation on beta-galactosidase were identified in the cotB regulatory region. The transcriptional activity of the distal and proximal regions' transcriptional activity was found to be additive, a property which was seen as beneficial to regulation by the cotB complete upstream region.

Whole-mount in situ hybridization of cell-type-specific mRNAs in Dictyostelium

Article Abstract:

The whole-mount in situ hybridization is a useful method for identifying the cells expressing the cell-type-specific mRNAs. The nonradioactive probes from cell-type-specific genes are hybridized to constant whole mounts formed at the mound, slug and culminant stages of Dictyostelium growth. Identical cellular patterns are obtained by labeling with probes from prespore gene and prestalk gene, and in strains having reporter constructs.
