Engineering of primary carbohydrate metabolism for increased production of actinorhodin in Streptomyces coelicolor

Article Abstract:

A study aims to determine the roles of key enzymes in central carbon metabolism in the context of increased production of antibiotics in Streptomyces coelicolor. Result reveal that under the conditions tested, glucose-6-phosphate dehydrogenase encoded by zwf2 plays a more important role than that encoded by zwf1 in determining the carbon flux to actinorhodin (Act), while the function of Pgm encoded by SCO7443 is not clearly understood.

Heterologous expression of novobiocin and clorobiocin biosynthetic gene clusters

Article Abstract:

An efficient method for the heterologous expression of biosynthetic gene clusters in different Streptomyces strains is described. Lambda-Red-mediated recombination in Escherichia coli is used for the introduction of integration functions into the cosmid backbones, which allowed subsequent site-specific integration into the genome of the heterologous host.

Region-specific transcriptional activity in the genome of Streptomyces coelicolor A3(2)

Article Abstract:

Researchers used subtractive hybridization between genome-specific DNA and total RNA and PCR to analyze gene expression in Streptomyces coelicolor A3(2) and the accessory linear plasmid SCP1. About 24 genes in the silent regions of S. coelicolor A3(2) chromosomes are expressed.
