Effect of inducible thrB expression of amino acid production in Corynebacterium lactofermentum ATCC 21799

Article Abstract:

Design of a plasmid, which facilitates separate transcriptional regulation of each gene, helps evaluate the influence of various enzyme activity ratios on metabolite accumulation in Corynebacterium lactofermentum ATCC 21799. Increase in homoserine kinase activity relative to homoserine dehydrogenase (HD) activity leads elimination of homoserine accumulation in the medium and significantly increases the final threonine titer. Feedback-insensitive HD enzyme regulates the carbon flux distribution at the DL-aspartic-beta-semialdehyde branchpoint.

A novel screening method for isolating exopolysaccharide-deficient mutants

Article Abstract:

A new screening process for detecting exopolysaccharide-deficient mutants that do not require Calcofluor binding was discovered. The technique may be applied for the isolation of mutants of Rhizobium meliloti that do not have normal production of either succinoglycan or galactoglucan. Exopolysaccharide-producing strains of R. meliloti were observed to exclude the lipophilic dye Sudan Black B, while exoB mutants which do not synthesize succinoglycan and galactoglucan readily accepted the dye.

Development and validation of Corynebacterium DNA microarrays

Article Abstract:

Researcher evaluate gene expression in Corynebacterium glutamicum using DNA microarrays. The results agreed with other methods used for this purpose. C. glutamicum is used industrially to make lysine and glutamic acid.
