Article Abstract:
A study was conducted to analyze the association between receptor tyrosine kinase activated signaling pathways and the transcriptional induction of immediate early genes. Global expression monitoring was utilized to identify 66 fibroblast immediate early genes induced by platelet-derived growth factor beta receptor signaling. Results indicated that diverse pathways promote overlapping effects on gene induction.

DNA probes revealed that PDGF activates inactive serine/threonine protein kinase encoded by Akt proto-oncogenes. The Akt activation was shown to be accompanied by mutations in tha Akt Pleckstrin domain and dependent on PDGFbeta tyrosines Y740 and Y751, which binds to phosphatidylinositol 3-kinase. Howver, activation was blocked by inhibitory proteins led by PI 3-kinase inhibitor wortmannin and other N17Ras.

The function of phosphoinositide 3-kinase (PI3K) in intracellular signaling has been underscored by its implication in a wide array of biological responses. One of the more recent findings indicates that PI3K is involved in providing cellular protection against programed cell death or apoptosis through a signaling pathway that involves the serine\threonine protein kinase Akt/PKB.
