Distribution of bacterial growth activity in flow-chamber biofilms

Article Abstract:

A novel reporter system was constructed for the monitoring of cellular growth activity. By constructing an expression cassette consisting of the novel green fluorescent protein gene expressed from an Escherichia coli rRNA promoter, the reporter system was derived, generating green fluorescence in fast-growing cells of both E coli and Pseudomonas putida. Results indicate that the dynamics of biofilm growth activity can be traced to the level of individual cells, cell clusters and microcolonies.

Effect of bacterial distribution and activity on conjugal gene transfer on the phylloplane of the bush bean (Phaseolus vulgaris)

Article Abstract:

A study was conducted to investigate the significance of bacterial distribution and metabolic activity to the conjugation on the phyloplane. The conjugal plasmid transfer was analyzed and the spatial distribution of metabolic pattern of the bacteria was examined. Results revealed that transconjugants can be observed in the junctures between epidermal cells and substomatal cavities and their distribution was similar to the distribution of other bacteria on nonsterile leaves.

New unstable variants of green fluorescent protein for studies of transient gene expression in bacteria

Article Abstract:

The green fluorescent protein (Gfp) from the jellyfish Aequorea victoria is a useful reporter in monitoring biological processes in situ because no additional substrate is required to express the green fluorescence. Short peptides were added at the carboxyl terminal of intact Gfp to generate unstable variants with variable half-lives. These variants can be utilized in the investigation of temporal gene expression in situ.
