Differentiation of Methanosaeta concilii and Methanosarcina barkeri in anaerobic mesophilic granular sludge by fluorescent in situ hybridization and confocal scanning laser microscopy

Article Abstract:

Methanosaeta concilii has been differentiated from Methanosarcina barkeri in anaerobic mesophilic granular sludge using fluorescent in situ hybridization and confocal scanning laser microscopic methods. Oligonucleotide probes were developed from genes coding for 16S ribosomal DNA to differentiate the two bacteria from mesophilic methanogens. The probes proved effective in the identification, localization and quantification of the specific relative abundance of Methanosaeta concilii and Methanosarcina barkeri.

In situ localization of Azospirillum brasilense in the rhizosphere of wheat with fluorescently labeled, rRNA-targeted oligonucleotide probes and scanning confocal laser microscopy

Article Abstract:

A combination of scanning confocal laser microscopy and fluorescently labeled oligonucleotide probes helps analyze the in situ localization of Azospirillum brasilense in wheat roots. Root hair zone has high levels of A. brasilense Sp7 and Wa3, whereas the interior of root cell has high levels of strain Sp245. In the rhizosphere, the physiologically active form is C-form of A. brasilense.

Effect of Octamethylcyclotetrasiloxane on methylation of bismuth by methanosarcina barkeri

Article Abstract:

A comparative study on the ionophores monensin and lasalocid which induce effect similar to those observed for Octamethylcyclotetrasiloxane (OMCTS) was conducted. It was suggested that the stimulation observed was mainly due to facilitated membrane permeation of the metal ion and stimulation of bismuth methylation by Methanosarcina barkeri was not specific for siloxane.
