Development of Techniques to genetically manipulate members of the genera Cytophaga, Flavobacterium, Flexibacter, and Sporocytophaga

Article Abstract:

Suicide vector pLYL03 that can replicate in Escherichia coli but fails to do so in Cytophaga, Flavobacterium, Flexibacter, and Sporocytophaga, is constructed. It carries transposon Tn4351, which causes transposon induced mutations in the host genome and confers erythromycin resistance. Presence of Tn4351 in the erythromycin resistant bacteria is detected by southern blots. Auxotrophic mutations of Flavobacterium is obtained after Tn4351 mutagenesis. The construction of a suicide vector, a selectable marker, and transposon can facilitate genetic work on these bacteria.

Effect of insertion site and metabolic load on the environmental fitness of a genetically modified Pseudomonas fluorescens isolate

Article Abstract:

There is a lack of empirical data concerning genetically modified microorganisms (GMMs) and their environmental fitness. In an effort to address this problem, a Pseudomonas fluorescens isolate was genetically modified for application in various environmental conditions. Upon analysis, fitness of the recombinant organism was found to be dependent on the environment. Metabolic load was also found to affect fitness. On the other hand, site of chromosomal insertion of marker genes had little effect on the fitness of the recombinant organisms.

Insecticidal properties of genetically engineered Baculoviruses expressing an insect juvenile hormone esterase gene

Article Abstract:

The gene coding for juvenile hormone esterase (jhe) was inserted into the genome of a Baculovirus containing a synthetic viral promoter. JHE was detected 12 hours after infection of Spondoptera frugiperda cells. Immunoblotting showed that the JHE produced was heterogenous, due probably to differential glycosylation. The recombinant gene was also expressed in vivo, however, no significant differences in weight, development or time of mortality was detected between larvae infected with jhe-expressing or -non-expressing virus.
