Determination of complement-mediated killing of bacteria by viability staining and bioluminescence

Article Abstract:

Escherichia coli and Bacillus subtilis strains were used to test the applicability of complement-mediated killing to both gram-negative and gram-positive bacteria and were monitored by flow cytometric, luminometric and conventional plate counting methods. The levels of bioluminescence emitted by B. subtilis and E. coli decreased with increasing serum concentration. All the three methods gave the same killing rate, suggesting that the bacteriolytic activity of serum complement can be measured rapidly and conveniently by using viability stains or bioluminescence.

Role of efflux pumps in adaptation and resistance of Listeria monocytogenes to benzalkonium chloride

Article Abstract:

The role of efflux pumps in adaptation of previously characterized isolates of Listeria monocytogenes that are either sensitive or resistant to benzalkonium chloride (BC) and also the role of plasmids in natural resistance of L. monocytogenes to sanitizers are investigated. The analysis has shown that not only the presence of the respective efflux pump genes but also their expression under different conditions should be examined to develop strategies for efficient control of L. monocytogenes.

Assessment of photodynamic destruction of Escherichia coli O157:H7 and Listeria monocytogenes by using ATP bioluminescence

Article Abstract:

The effects of photodestruction on Escherichia coli and Listeria monocytogenes cells are evaluated, using a bioluminescence method for detection of ATP leakage and a colony-forming assay. The time courses of photodestruction and intracellular ATP leakage were different for E. coli and L. monocytogenes and the results show that bioluminescent ATP-metry can be used for investigation of the first stages of bacterial photodestruction.
