Article Abstract:
Quantification of biofilm structures has been carried out using a novel computer program, COMSTAT. The program has ten features for quantifying three-dimensional biofilm image stacks. Single-species biofilms of each of four Pseudomonas strains were tagged with green fluorescent protein and grown in flow chambers with a defined minimal medium as substrate. The four strains show different modes of biofilm growth.

Article Abstract:
A general method for quantitative comparison of biofilm structures and assessment of experimental reproducibility between independent biofilm experiments is presented with Pseudomonas aeruginosa study an example. The method depends on study using a novel computer program, COMSTAT. Structural organization of microbial communities is influenced by many factors which can be studied in this way.

N-acylhomoserine lactones (AHL) have been detected in Pseudomonas aeruginosa-infected lungs of mice. This shows production and excretion of AHL molecules in vivo by the infecting organism. The view that expression of AHL molecules by P. aeruginosa during infection coincides with its pathogenesis has been supported. Green fluorescent protein (GFP)-expressing E. coli bacteria could be detected in the lung tissues after mice were challenged intratracheally with alginate beads having P. aeruginosa and E. coli and then killed on different days Pathogensis of P. aeruginosa is seen with expression of virulence factors, many controlled by two AHL-based quorum-sensing systems.
