Article Abstract:
A transcription elongation factor from HeLa cell nuclear extracts has been identified. In conjunction with the transcription inhibitor 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole (DRB) it causes pausing of RNA polymerase II (Pol II). The factor is called DRB sensitivity-inducing factor (DSIF). It has been purified and it is made up of 160-kD (p160) and 14-kD (p14) subunits. IT seems to associate with RNA Pol II and regulates its processivity in vitro and in vivo.

Article Abstract:
A study was conducted to analyze a transcription system that reconstitutes the sensitivity of a classic inhibitor of transcription elongation, DRB, in vitro. The system utilizes recombinant general transcription factors and DSIF. A factor called NELF represses pol II elongation and is counteracted by P-TEFb-dependent phosphorylation of the pol II CTD. Results indicated a molecular mechanism for DRB action and a new aspect of C-terminal domain phosphorylation.

Article Abstract:
A novel mechanism of attenuating the ER alpha-activity is described. It is shown that cofactor of BRCA1 (COBRA1), an integral subunit of the human negative elongation factor (N ELF), directly binds to ER alpha and represses ER alpha-mediated transcription.
