DNA amplification polymorphisms of the cultivated mushroom Agaricus bisporus

Article Abstract:

The genomes of commercial, wild-type and homokaryotic strains of Agaricus bisporus were analysed through the randomly amplified polymorphic DNA (RAPD) loci. Of 20 single 10-bp primers tested, 19 produced scorable markers. Analyses of the RAPD patterns showed that the method could differentiate between various strains of A. bisporus. In addition, RAPDS could identify haploid nuclear components of a heterokaryon, and verify artificially synthesized heterokaryons. The transfer of loci could be readily monitored using this technique. These results demonstrate the usefulness of RAPD analysis in strain fingerprinting and genetic studies in A. bisporus.

Article Abstract:

Morphological, molecular, and inbreeding features of mushroom belonging to section Arvenses demonstrate that they exhibit predominantly heterothallic breeding system. Data further point out that the breeding behavior favors both interbreeding and homokaryotic fruiting.
