Article Abstract:
The karyotype of the tamaraw was studied using RBG-banding methods and compared with those of the river and swamp cytotypes of domestic water buffalo. The tamaraw karyotype comprises six submetacentric and 16 acrocentric autosome pairs and X and Y chromosome. Chromosome 2 of the tamaraw had larger hybridization signals for the buffalo satellite 1 DNA, and it may have occurred due to relatively recent centromeric fusion.

Two types of satellite DNA sequences from the swamp-type buffalo and river-type buffalo were molecularly cloned and characterized, and the chromosomal distribution of the satellite DNA sequences was investigated, using fluorescence in situ hybridization (FISH). Centromere-specific satellite DNA was found in the interstitial region of the long arm of chromosome 1 of the swamp-type buffalo. The intensity of the hybridization signals with buffalo satellite II DNA included the Y chromosome.
