Article Abstract:
The first crystal structure of mitochondrial Complex II from porcine heart at 2.4 Armstrong resolutions and its complex structure with inhibitors 3-nitropropionate (NPA) and 2-thenoyltrifluoroacetone (TTFA) at 3.5 Armstrong resolutions are reported. Complex II is comprised of two hydrophilic proteins, flavoprotein (Fp) and iron-sulfur protein (Ip), and two transmembrane proteins (CybL and CybS), and the prosthetic groups required for electron transfer from succinate to ubiquinone.

Article Abstract:
A novel approach is employed to identify the 75 kDa subunit of complex I as a caspase substrate accessible to the intermembrane space of the mitochondria. It is found that mutation of the single, conserved caspase cleavage site leads to protection of mitochondrial functions and integrity during apoptosis.

A pathway for the import of proteins into the intermediate space (IMS) of mitochondria is described. The proteins, Erv1 and Mia40, function as a disulfide relay system that catalyzes the import of proteins into the IMS by an oxidative folding mechanism.
