Critical role for Atm in suppressing V(D)J (ital) recombination-driven thymic lymphoma

Article Abstract:

Atm has a critical role in suppressing V(D)J (ital) recombination-driven thymic lymphoma, as has been shown by introducing a RAG-1 deficiency into mice that are atm(super.-/-) in the presence of a TCR transgene. Chromosome translocations involving T cell receptor (TCR) loci have been found in tumors from Ataxia telangiectasia (AT) patients and in mouse ATM (super.-/-) thymoma. V(D)J (ital) recombination is probably involved in these malignancies.

Leukemia and lymphoma: A cost of doing business for adaptive immunity

Article Abstract:

A majority of B- and T-cell leukemias and lymphomas which are associated with chromosomal abnormalities that bear the hallmarks of aberrant V(D)J or class-switch recombination are described and the cost is genomic instability and its potential contribution to malignancy. Examples of B-cell lymphomas are also explained with point mutations in proto-oncogenes consistent with a role for somatic hypermutation (SHM).

Oncogenic Abl and Src tyrosine kinases elicit the ubiquitin-dependent degradation of target proteins through a Ras-independent pathway

Article Abstract:

Loss of Abi proteins, substrates of Abl kinases, appears to play a role in the progression of Bcr-Abl-positive leukemias. The destruction of Abi proteins involves a Ras-independent pathway, is triggered by activity of oncogenic Abl and Src tyrosine kinases, and depends on the ubiquitin-proteasome pathway.
