Construction of environmental DNA libraries in Escherichia coli and screening for the presence of genes conferring utilization of 4-hydroxybutyrate
Article Abstract:
Research was conducted to examine the potential of prepared libraries and to test the accessibility of the genetic diversity in an environment by direct cloning of environmental DNA. The direct lysis method was used to prepare the DNA while envivronmental libraries were built using Escherichia coli as the host. Results indicate that it is possible to prepare DNA libraries and to clone directly functional genes from environmental soil samples.

Construction and screening of metagenomic libraries derived from enrichment cultures: generation of a gene bank for genes conferring alcohol oxidoreductase activity on Escherichia coli
Article Abstract:
The DNA libraries from polyol-fermenting microorganisms are screened for the genes conferring a carbonyl compound-producing phenotype on Escherichia coli ECL707 upon incubation with short-chain polyols containing adjacent hydroxyl groups. The gene bank is constructed from the positive E. coli strains containing plasmids encoding nicotinamide-dependent dehydrogenase activity.
