Conservation of structure and mechanism between eukaryotic topoisomerase I and site-specific recombinases

Article Abstract:

Research was conducted to study the crystallization of the C-terminal catalytic domain Topo(81-314) and the determination of its structure. Topo(81-314) was determined in Escherichia coli BL21 cells under the influence of a T7 promoter. Protein expression was conducted by the infection of an 8 I culture with bacteriophage lambda CE6. Results showed that the C-terminal catalytic domain was characterized by a protease-sensitive region from residues 135-147.

Positional information within the Mu transposase tetramer: catalytic contributions of individual monomers

Article Abstract:

Research indicates that the reactions involved in Mu transposition are mediated by Mu transposase monomers contained in Mu protein tetramers. Reactions between a pair of Mu inactive variants show that the Mu monomers are contained in the transpositional complex serving as a factor for nucleoside splitting and in the transferring of DNA (Deoxiribonucleic acid) strands. MuA binding elements also contain both domain II for R1 and domain III alpha for R2.

The 3' enhancer region determines the B/T specificity and pro-B/pre-B specificity of immunoglobulin V(sub kappa)-J(sub kappa) joining

Article Abstract:

The immunoglobulin kappa gene 3' enhancer (E3') suppresses V(sub kappa)-J(sub kappa) recombination and restricts it to the pre-B developmental stage of B cells. The B/T specificity of V(sub kappa)-J(sub kappa) recombination is controled by the PU.1 binding domain in E3'. A mutation in the PU.1 domain causes recombination in both B and T cells. Deletion of E3' causes rearrangement of the kappa gene in the early pro-B stage of B cells.
