Article Abstract:
A study of the formation of recombinants at the time of Hfr X F- crosses of Escherichia coli by checking the integration of Hfr DNA into the circular chromosome of the recipient through unselected markers reveals that inheritance of the Hfr DNA in a single segment occurs in 60-70% of the recombinants in crosses that choose a marker positioned approximately 500 kb from the Hfr origin. When the markers are situated 700-2200 kb from the origin, the fraction of recombinants with multiple proximal markers accompanies such increase. Recombination efficiency decreases and the proximal exchange occurs closer to the selected marker following mutations of recB or recD plus recJ in the recipient.

Article Abstract:
An experimental system consisting of plasmid substrates fused to EcoRI sites at lambda phage arms was used to investigate the double-strand break (DSB)-stimulated plasmid recombination by the RecE pathway. The results showed that DSB could induce more than one type of recE-dependent recombination pathway in recB recC sbcA mutants. The type of recombination pathway depends on the location of the break with respect to homology.

Products and intermediates of RecE recombination pathway of Escherichia coli mutants were isolated and characterized after infection of recB recC sbcA mutants expressing EcoRI endonuclease with chimeric lambda phages. Heteroallelic recombinants were produced while some of the nonreplicative recombination products have heteroduplexes with a strong bias for paired strands ending 3' at the break.
