Combining the sibling disequilibrium test and transmission/disequilibrium test for multiallelic markers

Article Abstract:

The multiallelic sibling disequilibrium test (SDT) can be combined with the multiallelic transmission disequilibrium test (TDT). The component sign test described by Horvath and Laird in their description of the SDT can be extended to incorporate TDT data. A procedure is outlined for employing TDT data from families in which more than one child is affected or for cases in which genotyping is available for only one parent.

Power, mode of inheritance, and Type I error in LOD scores and affecteds-only methods: reply to Kruglyak

Article Abstract:

LOD scores represent a valid method of data analysis in linkage studies. It is misleading to suggest that nonparametric tests are purer or have more power than the relevant LOD-scores analysis. L. Kruglyak has raised issues concerning the terms 'nonparametric' and 'model free,' power and LOD scores, and the role of the true mode of inheritance.
