Article Abstract:
Two different O demethylation systems in lignin metabolism by Sphingomonas paucimobilis SYK-6 and their coexistence are discussed relative to cloning and sequencing of the lignin biphenyl-specific O-demethylase (LogX) gene. LigX is essential for DDVA O demethylation in SYK-6. One system is a tetrahydrofolate-dependent methyltransferase system and the other is a oxygenative demethylase one.

Article Abstract:
A second 5-carboxyvanillate (5CVA) decarboxylase gene, ligW2 was characterized and isolated and the roles played by both ligW and ligW2 in 5,5'-dehydrodivanillate (DDVA) catabolism were examined. Results indicate that both ligW and ligW2 contribute to 5CVA degradation, although ligW2 plays the more important role in the growth of SYK-6 cells on DDVA.

The Sphingomonas (Pseudomonas) paucimobilis gene involved in vanillate and syringate O demethylation was cloned and sequenced. Two mutants with altered dehydrodivanillic acid (DDVA) degradation pathways were isolated. Degradation of DDVA was not possible with the mutant strain NT-1, although syringate, vanillate and OH-DDVA was. Strain DC-49 could not degrade either vanillate or syringate, but could assimilate DDVA slowly. Only the ligH gene was found to complement both vanillate and syringate O demethylation of strain DC-49, but not in strain NT-1 by pDE20.
